There are machines that look similar to dial indicators that measure density of cane for you.
There are also tests you can do. In the Double Reed (IDRS Quarterly publication) a while back ago, there was someone who did an article about measuring cane density by:
1. Getting a narrow glass (like a chemistry test tube)
2. Filling each tube with a precise amount of water and measuring the height of the water.
3. Putting a piece of cane in the tube so that it is vertical.
4. Waiting for a certain amount of time, (1 or 2 hours) and measuring the amount of water absorbed, and the height of the water.
The softer the cane, the more water it absorbs.
