Starting from a blank, it takes me about 5 or 10 minutes to make a reed to the point that it is playable, but a little heavy. At that point I like to put it aside until the next day and spend a couple more minutes on it. If I have a rehearsal the next day, I'll finish it on site since the acoustics and temperature of the room will affect how the reed feels.
My best reeds are probably the ones where I start from a dry blank and do the two-step procedure above. Although I can tie-up a reed and make a playable reed in one step, these don't tend to last as long since cane soaked enough to tie on a staple behaves differently from cane that has been allowed to dry thoroughly at least once and then soaked just enough to scrape.
