Does having more wood in the middle of a barrel, somehow make the sound darker?

Yes, according to the barrel makers who make thicker walled barrels.
Maybe, but it depends on what you mean by "darker."

Without having 2 barrels dentical in every respect except wood thickness it's not easy to judge precisely. However I do find that some extra wood in the barrel tends to reduce the "brightness" a little.
I usually use an RC barrel on my Leblanc LL Bb for just this reason, it has virtually identical bore diameter and taper as the Leblanc barrel but about 2mm larger external diameter.
B&H usually chose realtively large diameter barrels on their 1010 and 926 range and Peter Eaton barrels again are quite large, typically about 35mm.
Many French barrels were 31-32mm however some of the monstrous ones I have seen pictured recently must be 36 - 40mm and I can't help thinking that this would start to deaden the tone (maybe thats what some people call "dark").

I read an article that the Tosca barrel was thicker for this reason. The Backun (appropriately named) 'Fatboy', must have some rational as to why it is so wide.
Just wondered if any experts had some real info on this?

The added mass imparts dimensional stability to some of the lighter woods (cocobolo, rosewood, etc) in the face of humidity and temperature changes. A barrel need not be overly massive, IMHO, to achieve this benefit.
As far as darker or brighter......that would depend on the wood, the internal topography, and (to quote G. Smith, the spirit of the reamer), and...oh yes, most importantly, how you play it.

Does a reverse taper (like moenig or Chadash) give a darker sound, or does this just give a more compact sound?
Have you done any tests to measure whether a thicker barrel gives more strength to the lower partials and/or reduces the strength of the upper partials?Or is it all at the mercy of the Grim reamer?

FWIW, I use the term "mass" rather than "width" to indicate the amount of material that comprises the barrel external to the bore.
Otherwise, people confuse "width" with diameter of either the bore or sockets. Likewise, the use of the word "volume" in this context can be confusing (amount of wood vs. volume of the bore column)
