I currently have a Yamaha Advantage YA 200AD Student Clarinet and am looking for a step up replacement. I have played a selmer signet and really enjoyed that and i have also heard good things about the Buffet E11 and the LeBlanc Bliss. Will I be able to afford a vintage one of these? Where should I look for them? A bit of background I am the 2nd chair high school clarinetist in the 2nd band at my school and I also play recreationally in a church youth ensemble.

The closer something is to your face, the more difference it makes. In my opinion, you'll do better by getting a handmade mouthpiece and barrel for your Yamaha. I've put my mouthpiece and barrel on a plastic Vito, and no one could tell the difference between that and my Buffet R13. Even I had trouble telling the difference.
Walter Grabner, Greg Smith and Clark Fobes offer matched mouthpieces and barrels. The combinations sell for around $500.

Keep in mind, Ken would likely sound good on almost any setup, but his advise is sound too. .

i have already replaced the mouthpiece with a Vandoren 5rv so would the mouthpiece replacement still apply? And what if i wanted a new horn and not just a new mouthpiece?

Vandoren machine-made mouthpieces are good for what they are. A handmade mouthpiece is in a different league. Matched with a handmade barrel, the improvement is not subtle.

Your mouthpiece should be fine. Yamaha and Buffet are probably your best choices for an intermediate clarinet. Try before you buy. Pay particular attention to the upper clarion and altissimo.

Since you're already accustomed to the ergonomics of the Yamaha range, if you're happy with it why not go for something a bit higher in the Yamaha hierarchy?
A YCL450 or 650 or similar.
