I am due to play Tristan and Isolde and would like to prepare as well as I can before attending rehearsals.
Does anyone know the Opera well, and has any insight into the extracts most important to learn and prepare.

There aren't really any big clarinet solos in Tristan. (There is a huge bass clarinet part in act 2, when King Mark discovers that Tristan has betrayed him).
To prepare yourself well, it would be worth looking at the score, especially in the Prelude. Play though the chords on a piano, looking specifically at the exposed woodwind sections. Analyse the harmony. Figure out what your function is in the different chords to help you work out how best tune with the other players. If you have time in the rehearsals, get together with the other woodwind players and build up the chords- starting with root, then fifth, 3rd and then 7th. Sometimes it's hard to work out what the function of the chord is, and sometimes it's not easy to immediately hear what the harmony is because it's quite chromatic. But once you've worked it out and tried it with the others, it becomes much easier to play in tune.
