I think it's the length and the thickness of the shaft (the part you don't play on). The back of the reed is really fat compared to other reeds, so it vibrates differently than the regular VanDorens.

Because the V-12s have thicker blanks, the material that forms the tip comes from deeper within the tube of cane. In general, the xylems (the part the water passes through to supply the leaves of the plant) are concentrated just beneath the bark of the cane.

Therefore, V-12s will have fewer and thinner xylems, which are the dark streaks you see when you hold a reed up to the light. Thus, even though V-12s are thicker than the "traditional" Vandorens, they run slightly softer.

There's so much variability in Vandorens that I can't really tell whether this makes a lot of difference, but a lot of people think it does, so it's worthwhile experimenting.
The Gonzalez/Davie Cane models, which I've been playing recently, are available in similar designs -- the FOFs are made with a standard thickness blank, and the Mozarts have a thick one.
