Most likely, as many have suggested, it's mold caused by moisture that isn't
drying in whatever storage conditions are being used.

Of course, there is another outside possibility (I don't remember if the
student was a boy or a girl or how old) that some kind of lipstick/chapstick
or other lip dressing is responsible - usually a color of some kind, but if
it catches and holds enough dust, it could appear dark enough to be seen as
black. Doesn't seem likely from your description, but none of us can see the
spot. Is the spot where the lips contact the reed or somewhere else?
