Jim,
I think the opera you are thinking
of could be "Le Toreador" by
Adolphe Adam. Half way through there
is a trio in which Don Belflor and Tracolin
join Coraline in a trio (music lesson) "Ah!
vous dirai-je, maman" which we know as
"Twinkle, twinkle etc".
This ends with a flute playing the tune while
the singers do their respective variations.

There is the long Music lesson in Donizetti's
"La Fille du Regiment" but this isn't "Ah! vous
dirai etc...."

What opera has a "music lesson" on stage where the soprano and a flute do
essentially a duet. It seems that the "Twinkle, Twinkle, Little Star"
melody is involved.
