Based on this suggestion, I took the key off, ran a pipe cleaner up the
register tube.

Bingo. Even though I could blow through the register hole, and of course
other notes sounded in all registers, there must have been something in
there that stopped this note from sounding. It still does not sound
easily, but it does sound, and I think that the rest will have to come
with practice. This was barely a clog, I suspect, but it was enough to
interfere with the particular note.
