Hi Frank -

I personally find that soaking reeds too long water loggs them and they
blow out quickly. I DO like soaking my reeds in water over the mouth
method, but use the mouth method out of convienience more often.
Typically, I soak the entire reed (tip to heel) in water for about 20
seconds. Immersing the reed in my mouth, it takes just over a minute to
equally wet the reed (since saliva is thicker, it absorbs slower than
water does). I don't detect a notable change in tone quality between the
two, to be honest, and I use nearly the exact same setup as use (V12s at
4.5, a Genussa GE, and Bay lig).

Of course, that's just my experience. (Alto/Bass/Tenor/Bari/Contra reeds,
of course, take much more time.)
