Lorne G. Buick, Nathaniel Johnson and David Pino are all correct. I use the
pitch produced by the mouthpiece as a guide for my students also. For me,
that pitch can vary from C (concert) to a very flat C sharp (concert)
depending on the mouthpiece I use. I also use the mouthpiece and barrel with
students, mostly when I can't stand their mouthpiece sound any more. :-)

Dr. Pino gives very specific instructions in his book about producing the
"second F-sharp above middle C on the piano". His instructions in the section
"Embouchure Formation" are to "remove both the mouthpiece and the barrel as
one unit".
